Abstract: This study developed an adjuvant-type lipid. By replacing a portion of the ionizable lipid in LNP, it not only enhanced the mRNA delivery efficiency but also endowed LNP with Toll-like receptor 7/8 (TLR7/8) agonistic activity, thereby significantly boosting the immune response of mRNA vaccines while maintaining good tolerability.

This study presents a novel strategy for the synthesis of degradable branched lipids based on a "one-pot" approach. By in-situ construction, it enables the rapid and combinatorial expansion of lipid structures, which not only boosts the mRNA delivery efficiency by up to three orders of magnitude but also significantly simplifies the synthesis process, greatly enhancing production efficiency and effectively reducing the costs of synthesis and screening.
2024.7.9 nature chemistry
This study developed a one-pot sequential multi-component reaction strategy that enables the rapid and efficient synthesis of structurally diverse biodegradable lipid molecules at room temperature. This method significantly simplifies the traditional lipid synthesis process and achieves high-throughput and low-cost combinatorial construction. The lipid library constructed based on this strategy reveals key structural-functional relationships and significantly improves mRNA delivery efficiency. This synthesis platform provides important support for the discovery and large-scale application of new-generation efficient LNP delivery materials.

This study achieved the directed chemical evolution of 3-alkynyl-ionic lipids (A3-lipids) through iterative chemical derivatization and combinatorial chemistry strategies, using the amine-aldol-alkyne coupling reaction. After five rounds of iterative optimization, a variety of asymmetric A3 lipids with high delivery activity and good biodegradability were rapidly obtained, and their performance could be comparable to or even superior to that of the benchmark lipids. The established key structure-activity relationship on this basis provided important guidance for accelerating the development of the next-generation mRNA-LNP delivery system.
